      豬輪狀病毒GZAS2020株與NX株VP7蛋白的結構預測和抗原表位分析

      為了解豬輪狀病毒(PoRV)VP7蛋白的生物信息學信息,運用Expasy-ProParam、YinOYang-1.2、TMHMM-2.0、ABCpred和IEDB等軟件,預測分析PoRV臨床分離株GZAS2020和疫苗株NX VP7蛋白的基本理化性質、結構功能和B細胞抗原表位差異。結果顯示:兩種VP7蛋白均屬于穩定蛋白、親水性蛋白、跨膜蛋白和非分泌性蛋白;分離株與疫苗株的O糖基化位點數分別為55和62,N糖基化位點數均為1,磷酸化位點數分別為29和30;亞細胞定位均位于質膜,均以α-螺旋和無規則卷曲為主要結構,延伸和β-轉角貫穿其中;GZAS2020株的B細胞抗原表位位于67~72 aa(PYANSTT)、97~100 aa(KWTE)、176~179 aa(QQTD)、276~283 aa(TTAPQTER),NX株位于96~99 aa(TKWK)、176~182 aa(QQTDEAN)、274~279 aa(DPTTTP)、310~317 aa(MSKRSRSL),兩者在97~100、176~179、276~279 aa處存在相同的B細胞抗原表位,但GZAS2020株在無規則卷曲位置(67~72 aa)存在獨特的B細胞抗原表位。以上結果說明,疫苗株NX與貴州省流行株GZAS2020在主要抗原蛋白上存在差異。

      Comparison of Sequencing Results between PRRSV ORF5 Amplification Products and Its Cloning

      In order to learn the bioinformatics information of VP7 protein of porcine rotavirus(PoRV),the differences of general physicochemical properties,structural functions and B-cell epitopes of the clinical isolate GZAS2020 and the vaccine strain NX VP7 protein were predicted and analyzed by using Expasy-ProParam,YinOYang-1.2,TMHMM-2.0,ABCpred,IEDB and other software. The results showed that both the VP7 proteins belonged to stable proteins,hydrophilic proteins,transmembrane proteins and non-secretory proteins;the number of O-glycosylation sites in GZAS2020 and NX were 55 and 62,respectively,that of N-glycosylation site was 1,and that of phosphorylation sites was 29 and 30,respectively;all subcellular localization was located in the plasma membrane,with main structure of α-helix and irregular coil,through which the extension and β-turn run;the B-cell epitopes of the GZAS2020 strain were located at 67-72 aa(PYANSTT),97-100 aa(KWTE),176-179 aa(QQTD)and 276-283 aa(TTAPQTER),while those of NX were at 96-99 aa(TKWK),176-182 aa(QQTDEAN),274-279 aa(DPTTTP)and 310-317 aa(MSKRSRSL),both of which were with same B-cell epitopes at 97-100,176-179 and 276-279 aa,but unique B-cell epitopes were observed in the former at the position with irregular coil. It was concluded that the major antigen proteins of NX was different from those of GZAS2020 prevalent in Guizhou Province.
